Le risque thermique : caractérisation des environnements thermiques des 3 sites atliers de MOREST en lien avec les mortalités observées sur le terrain ArchiMer
Ropert, Michel; Rauflet, Fabienne; Soletchnik, Patrick; Geairon, Philippe; Bedier, Edouard; Bouget, Jean-francois.
Temperature affects undoubtedly the 3 working sites where, systematically, the observation of the first abnormal mortalities coincides with a water temperature of around 19°C. The nature of the processes implied could have a "cumulative" effect, affecting the physiology (this phenomenon has been observed with other marine species) or a "threshold" effect, acting only as a trigger. The question has not been answered yet. The kinetics of the thermic evolution around the mortality period has to be studied in depth in order to understand if this temperature acts more as an "associated factor", during a chronic stress period, or during an acute stress, as suggested by the "on-bottom/off bottom" mortality model of the Marennes Oléron Bay [12].

Leukocyte profile of the helmeted manakin, Antilophia galeata (Passeriformes: Pipridae) in a Cerrado forest fragment Rev. Bras. Zool.
Ribeiro,Paulo Vitor Alves; Baesse,Camilla Queiroz; Cury,Márcia Cristina; Melo,Celine de.
ABSTRACT Changes in the amounts and proportions of leukocytes, known as leucocyte profiles, have been documented for several bird species and have been used to measure stress levels in these animals. The present work ascertained the biological and ecological attributes that influence the leukocyte profile of Antilophia galeata (Lichtenstein, 1823), the helmeted manakin. This species has been deemed useful in ecological studies because it responds to environmental changes. Blood samples drawn from 89 individuals of A. galeata captured in a Cerrado forest fragment were subjected to analysis under optical microscopy to identify and quantify leukocytes and micronuclei. The number of lymphocytes was greater for males, non-reproductive individuals and...

Lipid peroxidation and total radical-trapping potential of the lungs of rats submitted to chronic and sub-chronic stress BJMBR
Torres,R.L.; Torres,I.L.S.; Gamaro,G.D.; Fontella,F.U.; Silveira,P.P.; Moreira,J.S.R.; Lacerda,M.; Amoretti,J.R.; Rech,D.; Dalmaz,C.; Belló,A.A..
Exposure to stress induces a cluster of physiological and behavioral changes in an effort to maintain the homeostasis of the organism. Long-term exposure to stress, however, has detrimental effects on several cell functions such as the impairment of antioxidant defenses leading to oxidative damage. Oxidative stress is a central feature of many diseases. The lungs are particularly susceptible to lesions by free radicals and pulmonary antioxidant defenses are extensively distributed and include both enzymatic and non-enzymatic systems. The aim of the present study was to determine lipid peroxidation and total radical-trapping potential (TRAP) changes in lungs of rats submitted to different models of chronic stress. Adult male Wistar rats weighing 180-230 g...

Measuring cortisol, the major stress hormone in fishes ArchiMer
Sadoul, Bastien; Geffroy, Benjamin.
Stress in teleosts is an increasingly studied topic because of its interaction with growth, reproduction, immune system and ultimately fitness of the animal. Whether it is for evaluating welfare in aquaculture, adaptive capacities in fish ecology, or to investigate effects of human‐induced rapid environmental change, new experimental methods to describe stress physiology in captive or wild fish have flourished. Cortisol has proven to be a reliable indicator of stress and is considered the major stress hormone. Initially principally measured in blood, cortisol measurement methods are now evolving towards lower invasiveness and to allow repeated measurements over time. We present an overview of recent achievements in the field of cortisol measurement in...

Metabolic responses and β-carotene production by the unicellular green alga Dunaliella salina exposed to leaf extracts Acta Botanica
Einali,Alireza; Mazang-Ghasemi,Sahar; Valizadeh,Jafar; Noorozifar,Meisam.
ABSTRACT The present work investigated the effects of aqueous extracts of eucalyptus ( Eucalyptus globulus) and elderberry ( Sambucus ebulus) leaves on β-carotene productivity in Dunaliella salina, a green microalga. Leaf extracts from eucalyptus have greater amounts of phenolics and flavonoids, as well as greater ferric reducing antioxidant potential than elderberry. The extracts of both species greatly inhibited growth of algal suspensions. However, chlorophyll and β-carotene concentration increased in cells treated with leaf extracts, and the highest values were detected in 1 % eucalyptus and 2 % elderberry extracts. Fresh weight, total sugar, and protein content significantly increased following exposure of cells to different doses of leaf extracts....

Molecular cloning and expression analysis of 12-oxophytodienoate reductase cDNA by wounding in Solanum tuberosum Electron. J. Biotechnol.
Díaz,Mauricio; Polanco,Victor; Ramírez,Ingrid; Peña-Cortés,Hugo.
Jasmonic acid (JA) and 12-oxophytodienoic acid (OPDA) are signal molecules involved in the stress and defense responses in plants. A full-length cDNA clon of OPR3 encoding 12-oxophytodienoate reductase 3, key enzyme involved in the biosynthesis of JA from linolenic acid was obtained from a Solanum tuberosum cDNA library. Sequence analysis showed that OPR3 encoded a polypeptide of 400 amino acids with a predicted molecular mass of 43.9 kDa and pI of 7.72. The deduced amino acid sequence of OPR3 showed high similarities to other 12-oxophytodienoate reductases. A peroxisomal signal sequence indicates OPR3 probable location in peroxisome. Levels of OPR3 mRNA accumulated in potato leaves reaching maximum levels within 1 hr of mechanical wounding. Elevated...

Mycobacterium avium subsp. Paratuberculosis and the expression of selected virulence and pathogenesis genes in response to 6°c, 65°c and ph 2.0 BJM
Pribylova,Radka; Kralik,Petr; Donnelly,Neysan; Matiasovic,Jan; Pavlik,Ivo.
The aim of this work was to study the expression of selected Mycobacterium avium subsp. paratuberculosis (MAP) genes connected with MAP virulence, adhesion and stress response. The temperature of 6°C and 65°C were chosen with regard to the food industry, storage conditions (refrigerator) and low-temperature pasteurization. A pH of 2.0, using lactic acid, was selected to mimic the natural environment of the stomach. Expression of selected genes was studied using real time reverse transcription PCR on three different MAP isolates. MAP isolates were chosen according to the number of their preceding cultivations. While isolates 8672 and 8819 were previously cultivated only once, MAP isolate 12146 went through four passages. Different expression profiles were...

Neonatal handling and the expression of immunoreactivity to tyrosine hydroxylase in the hypothalamus of adult male rats BJMBR
Hermel,E.E.S.; Severino,G.S.; Cecconello,A.L.; Pereira,F.M.; Sanvitto,G.L.; Lucion,A.B..
Neonatal handling has long-lasting effects on behavior and stress reactivity. The purpose of the present study was to investigate the effect of neonatal handling on the number of dopaminergic neurons in the hypothalamic nuclei of adult male rats as part of a series of studies that could explain the long-lasting effects of neonatal stimulation. Two groups of Wistar rats were studied: nonhandled (pups were left undisturbed, control) and handled (pups were handled for 1 min once a day during the first 10 days of life). At 75-80 days, the males were anesthetized and the brains were processed for immunohistochemistry. An anti-tyrosine hydroxylase antibody and the avidin-biotin-peroxidase method were used. Tyrosine hydroxylase-immunoreactive (TH-IR) neurons were...

Neonatal handling induces anovulatory estrous cycles in rats BJMBR
Gomes,C.M.; Frantz,P.J.; Sanvitto,G.L.; Anselmo-Franci,J.A.; Lucion,A.B..
Since previous work has shown that stimulation early in life decreases sexual receptiveness as measured by the female lordosis quotient, we suggested that neonatal handling could affect the function of the hypothalamus-pituitary-gonadal axis. The effects of neonatal handling on the estrous cycle and ovulation were analyzed in adult rats. Two groups of animals were studied: intact (no manipulation, N = 10) and handled (N = 11). Pups were either handled daily for 1 min during the first 10 days of life or left undisturbed. At the age of 90 days, a vaginal smear was collected daily at 9:00 a.m. and analyzed for 29 days; at 9:00 a.m. on the day of estrus, animals were anesthetized with thiopental (40 mg/kg, ip), the ovaries were removed and the oviduct was...

Neuroethologic differences in sleep deprivation induced by the single- and multiple-platform methods BJMBR
Medeiros,R.; Lenneberg-Hoshino,C.; Hoshino,K.; Tufik,S..
It has been proposed that the multiple-platform method (MP) for desynchronized sleep (DS) deprivation eliminates the stress induced by social isolation and by the restriction of locomotion in the single-platform (SP) method. MP, however, induces a higher increase in plasma corticosterone and ACTH levels than SP. Since deprivation is of heuristic value to identify the functional role of this state of sleep, the objective of the present study was to determine the behavioral differences exhibited by rats during sleep deprivation induced by these two methods. All behavioral patterns exhibited by a group of 7 albino male Wistar rats submitted to 4 days of sleep deprivation by the MP method (15 platforms, spaced 150 mm apart) and by 7 other rats submitted to...
